Facilities and Major Equipment 
The proposed study mainly involves animal surgery, gait analysis, high resolution ultrasound imaging, 
tissue  processing  and  histology,  biomechanical  test,  and  fabrication  of  biomaterials  with  in  vitro 
characterization. 
In Hong Kong, the Lee Hysan (LHS) Clinical Laboratory in Prince of Wales Hospital provides a full gear 
of research facilities including tissue processing equipment, sectioning equipment (A) and various equipment 
for image analysis (i.e., motorized platform microscope (Leica) and image analysis software (Image Pro). The 
animal surgery will be performed in the Animal operation theatre located in the Li Ka Shing Institute of Health 
Sciences (LKSIHS), Faculty of Medicine, Prince of Wales Hospital. The High resolution ultrasound imaging 
machine (Visulasonics) (B) is also located in one  of the animal operation theatres. Gait capture using the 
Catwalk system (Noldus Technology) (C) will be performed in Animal Behavior Laboratory located in the same 
building. Biomechanical test will be performed using the universal material testing machine (D) (Hounsfield 
H25K-S, Tinius Olsen Ltd) for biomechanical study and it is located in the Musculoskeletal laboratory of the 
Department of O&T in LKSIHS. 
In Taiwan, the fabrication of mPEG-PLGA-Box block copolymer will be carried out in the Laboratory of 
Biomaterials and Tissue Engineering, which is located at Cardinal Tien Memorial Building in Fu Ren Catholic 
University in Taiwan. Facilities for biomaterial research such as lab for chemical synthesis, lyopholizer, NMR, 
HPLC, spectrophotometer, rheometer etc are available in the biomaterial lab.

This proposal will investigated the effect of
micelle-encapsulated quercetin on knee OA via
intra-articular injection. Although the PI's
proposal on GHK-Cu bioactive coating also
employs the use of micelle-based delivery
system, the type of polymer is different between
the PI's proposal (mPEG-PLGA-Box block
copolymer) and Prof. Hung’s proposal
(mPEG5000CPP). Moreover, GHK-loaded
micelle in the PI project will be a temperature-
sensitive gel that will form semi-rigid gel to
coat on the tendon graft; while in Prof. Hung's
proposal, the drug-loaded micelle will be
injected as suspension.

We have recieved a 1 year GRF grant working on related research topic, started from
2012Jan to 2012 Dec. The current research proposal is a continuation of this funded
project:
PI: Chan KM; CI: Yung Shu-Hang Patrick, Fu Sai-Chuen

Project Title:
A novel approach to tissue engineering in ACL surgery and healing -
accelerating tissue remodeling by GHK tripeptide
Nature of Project:
We proposed to investigate the effectiveness of intra-articular injection of GHK-Cu to
promote graft healing in a rat model of ACL reconstruction. The project was funded for 1
year with 320000 HKD. Our preliminary findings showed that repeated injections of
GHK-Cu can improve restoration of knee AP laxity but graft deterioration was still
evident. Therefore we propose to investigate a controlled delivery system for GHK-Cu
with a bioactive coating on tendon graft in the present proposal. We speculate that
controlled release of GHK-Cu and the protective layer can further improve the healing
outcomes in ACLR. The funded project is still on-going and expected to be completed by
Dec 2012.
We also received a GRF grant working on ACL reconstruction in rat model in the
2012/2013 exercise:

Project Title:
A Novel Tendon-Derived Stem Cell (TDSC) Sheet for the Promotion of Healing of
Anterior Cruciate Ligament (ACL) Graft – An Animal Study
Nature of Project:
This project is newly granted to investigate the possibility to use tendon-derived stem
cells to promote graft incorporation in ACLR. Although the same animal model is used,
this proposal is significantly different from our GRF proposal. The current GRF propsoal
will not work on stem cells, but focus on the development of controlled delivery system for
bioactive agents to promote graft healing in ACLR.
