Objective: Tendon-derived stem cells or tendon progenitor cells (TPC) are recently under 
extensitve investigation, and they may play important roles in tendon healing. It is not sure 
how  biological  environment  at  early  healing  stages  affect  recruitment  of  TPC.  As 
over-production of  reactive oxygen species (ROS) during inflammatory phase of  healing 
process can cause excessive tissue fibrosis, it is possible that recruitment of TPC relative to 
scarring fibroblasts is affected by oxidative challenge. In the present study, we investigated 
the effects of hydrogen peroxide on the clonogenicity of tendon progenitor cells (TPC). 
Methods:  TPC  were  isolated  from  rat  patellar  tendons  by  digestion  with  3  mg/ml 
collagenase. Colony-forming assay was performed at passage 0. Rat TPC were seeded at 50 
cells/cm
2
in control medium or test media (with  2, 10 or 50 uM H
2
O
2
). The cells were 
cultured for 7 days, fixed with ethanol and stained with 0.5% crystal violet for counting of 
colony-forming  units  (CFU).  CFU  with  higher  cell  density  (dense  clone  with 
cobblestone-shaped cells, > 250 cells/ mm
2
) and lower cell density (diffuse clones with 
fibroblast-like cells, < 250 cells/mm
2
) were counted with the help of ImagePro 6.2 software. 
Immunocytochemical staining for tenomodulin was employed to detect TPC-containing CFU. 
Results: We found that the total number of CFU was significantly decreased in the presence 
of  50  uM H
2
O
2
as compare to  control  (p<0.05). The average size  of the  CFU was not 
significantly different. Percentage of dense CFU was decreased but the percentage of diffuse 
CFU  was  increased  by  H
2
O
(p<0.05).  Almost  all  dense  CFU  were  immunopositive  for 
tenomodulin but diffuse CFU were negative for tendomodulin staining. 
Conclusion: The data suggest that oxidative condition may not favor recruitment of TPC 
(presumably for tendon regeneration), while fibroblastic cells (presumably for non-specific 
tissue repair) are preferred under oxidative conditions. It may help to explain our previous 
findings that anti-oxidant supplementation (vitamin C) at early healing stage can improve 
mechanical restoration of healing tendons. As chronic tendinopathy also involve oxidative 
stress, further studies on redox modulation on TPC may help to delineate the failed tendon 
healing responses in tendinopathy.
